You should really treat this part of your game more like long term planning. If you have a Banner for a Champ that is a big part of your team then yeah go ahead and Ascend him. So the real question is: Should you delay 6 Starring a Champ for this Mission if you're not there yet? I would say the answer to this question is "It depends." So the main reason you 6 Star Ascend is to get a Banner online. This is more of a public service announcement since I put a good amount of time researching stuff for Raid and I still missed this very annoying bump in the road.
